PROCUREMENT, RECEIPT, PREPARATION, STERILIZATION AND STORAGE OF MEDIA FOR MICROBIOLOGICAL ANALYSIS

  1. OBJECTIVE:

This SOP Describes the Procedure for Procurement Receipt Preparation Sterilization and Storage of Media for Microbiological Analysis.

  • SCOPE:

This SOP is Applicable to the Procedure for Procurement Receipt Preparation Sterilization and Storage of Media for Microbiological Analysis.

  • PROCEDURE
    • Receipt and Storage of Dehydrated Media:-
      • All required dehydrated media shall be procured from the Hi-media Labs. Ltd. Or any other reliable source.
      • On receipt of the media, it should be checked for their labeling details such as ingredients, media code number, use before date etc.
      • The date of receipt and other detailed shall be entered in the Media stock register & container should be labeled with detail of “Received on” date.
      • The dehydrated media are highly hygroscopic and must be stored in a cool dry place away from bright light or as instructions given on the label of the pack.
      • Container “opened” on date shall be mentioned on the container label, & recorded in the Media Stock in Format
    • Preparation of media :-  
      • Carefully read the instructions given on the label of the pack & check its Lot No. Ensure that the media is not deteriorated physically and check the use before date is not over.
      • For rehydration, use clean, undamaged glassware and Purified water & follow the directions given on the label for preparation of media.
      • Place appropriate weighed amount of medium in a clean dry flask add part of the required amount of Purified water. Swirl to dissolve and then add the remaining water from the side of container. For complete dissolving use hot plate/ heating mantle taking care to      avoid excessive heating or scorching of the medium.
      • Check the pH of the medium at room temperature & it should be within the range         specified on the label. (Adjust the pH if necessary by adding 1 N or 0.1 N HCl or NaOH solutions.)
      • Dispense completely dissolved medium either into conical flask or test tube depending on the further requirement. In case of conical flask & sampling bottles, close its mouth with non-absorbent cotton plug wrapped with butter paper or aluminum foil. In case of test tubes use caps or non-absorbent cotton plug wrapped with aluminum foil.
      • For autoclavable medias sterilie the prepared media by autoclaving at 15 psi pressure at 121°C for 15-20 minutes or per instruction available on media container. Enter the load details in the autoclave usage log card.
      • Assign a Lot Number to each set of prepared Media with “AbbrYYMMDD”.
      • Abbr is expressed abbreviation of media as per format no. F/MB-026-04.
      • YY is expressed year.
      • MM is expressed month
      • DD is expressed date.
      • Record the media preparation in the Media Preparation & reconciliation record as per Format.
      • Sterile agar media should be poured into plates at about 45-50° C to avoid the formation of condensed water in the lids of petri dish. The medium should be mixed thoroughly without bubble formation and aseptically poured into sterile petri plates.
      • Each media used record is maintained in Media Usage Record.
      • Expressed the abbreviation of media as per Format.
      • Preincubated the plate at 30 – 350C for 18 -24 hours. Maintain the record in Format.
