SOP for operation & calibration of TLC Cabinet

  1. OBJECTIVE:
to lay down a procedure for Operation & Calibration of TLC Cabinet.
  • SCOPE:
This procedure is applicable for procedure for Operation & Calibration of TLC Cabinet.

  • PROCEDURE:
    • Operating Procedure
      • Check the calibration status.
      • Develop the chromatogram as per the procedure specified in the individual monograph.
      • Mark the solvent front.
      • Dry the plate in air or as per the procedure.
      • Treat the developed plate if specified in the monograph as per the procedure given in the monograph.
      • Check the UV chamber for any unwanted things and remove them. Place the plate before the lamp in the chamber.
      • Switch ‘ON’ the mains.
      • Select the lamp of specified wave length (emission) by pressing the switch (red in colour) towards the wavelength indicated on the board.
      • View through the glass provided on the top of the chamber.
      • Mark the spots immediately.
      • Switch ‘OFF’ the power.
      • Remove the plate.

Calculate the Rf value by the formula

Rf = Distance traveled by the substance (spot) / Distance traveled by the solvent

Calibration:

For 254 nm (For Short Wavelength)Prepare a 0.04 % w/v solution of Sodium Salicylate in ethanol (95%).

Weight 40 mg of Sodium Salicylate and dilute to 100 ml of ethanol (95%).Apply 5µl of this solution on silica gel G with a diameter of about 5mm and examine visually.

The distance between the lamp and the plate under examination used in Pharmacopeia test should not exceed the distance used to carry out the above test.

The lamp should be capable of revealing without doubt a standard spot of Sodium Salicylate.

For 365 nm (For Long Wavelength)Prepare a 0.2 % w/v solution of Sodium Salicylate in ethanol (95%).

Weight 200 mg of Sodium Salicylate and dilute to 100 ml of ethanol (95%).

Apply 5µl of this solution on silica gel G with a diameter of about 5mm and examine visually

The lamp should be capable of revealing without doubt a standard spot of Sodium Salicylate.  Maintain the calibration Record.
