1.0 OBJECTIVE:
This SOP describes the procedure for operation and calibration of colony counter.
2.0 SCOPE:
This SOP is applicable to the procedure for operation and calibration of colony counter.
3.0 RESPONSIBILITY:
Microbiologist
4.0 ACCOUNTABILITY:
QC Manager.
5.0 PROCEDURE:
5.1 Operating Procedure:
- Ensure that the instrument is clean and suitable for use.
- Ensure that the magnifying lens is fixed on its appropriate plate.
- Ensure that the marker is connected to either right or left side of the plate.
- Switch on the mains.
- Switch on the power button of the instrument. The internal light will be flashed “ON” & the digital screen will show “0000” figure in red.
- Now press the plate containing colonies to be counted.
- Now, mark each & every colony one after by other by pressing the tip of the marker.
- The number of colonies will automatically get display on the screen can be set to”0000” by pressing of marker.
- If any correction in current counting of colony; correct the count by Count Correction key.
- During the counting of colony, hold the counting by press the Hold key.
- After completion of counting the colony, for next colony counting press the Reset Key.
- After completion the work, switch ‘OFF’ the Instrument.
- Maintain the usage record of colony counter .
5.2 Calibration:
- Operate the instrument as per the SOP & press the tip of the marker 10 times, 50 times & 100 times one by one then check the number displayed on the screen. This Method applied minimum three times after Reset the instruments. And record maintain .
- Frequency: Every Month.

Trainees : Staff of the microbiology departments
