PROCEDURE FOR OPERATION & CALIBRATION OF pH METER

1.0 OBJECTIVE:

To lay down a procedure for Operation & Calibration of pH meter.

2.0 SCOPE:

This procedure is applicable for procedure for Operation & Calibration of pH meter .

3.0 RESPONSIBILITY:

Microbiologist

4.0 ACCOUNTABILITY:

QC Manager / Microbiology In charge

 5.0 PROCEDURE:

5.1 Operation Procedure:

  • Ensure that the instrument is clean and suitable for use.
  • Ensure the calibration of instrument is done.
  • Switch ‘ON’ the mains.
  • Put the electrode in sample. Set the temperature by knob about 25 centigrade and check the pH by pH/S/mV Knob.
  • Display the pH in 0.00, note the pH.
  • Maintain its usage log book .

5.2 Calibration Procedure:

  • Ensure that the instrument is clean and suitable for use.
  • Switch ‘ON’ the mains.
  • Wash the pH electrode with purified water 2-3 times.
  • Deep the pH electrode in buffer solution pH 4.00 and operated by Step no. 5.1.4 to 5.1.6.
  • Check the pH of buffer solution pH 4.00; note down the pH in format of calibration of pH meter.
  • After check the pH of buffer solution pH 4.00; remove the pH electrode and wash with purified water 2 – 3 times. And then deep the pH electrode in buffer solution pH 7.00 for observation.
  • And after check the pH of buffer solution pH 7.00; remove the electrode and wash with purified water.
  • Deep the electrode in buffer solution pH 9.20 and observed the pH.
  • After completion of manual calibration check the pH 4.00, 7.00 & 9.20 one by one fill the Format .
  • Prepared the ph buffer solution as per Format & affix the label as per Format .
  • After use of tablet / capsule of pH buffer; note down in Format .

6.0 TRAINING:
